Palladium-catalyzed phosphine-free direct C-H arylation of benzothiophenes and benzofurans involving MIDA boronates

With high regioselectivity, a series of benzoheterocyclic compounds were synthesized via palladiium-catalyzed phosphine-free C-H arylation of benzothiophenes/benzofurans with aryl MIDA boronates at 30-50 C in moderate to excellent yields. MIDA boronates were used in C-H arylation of heterocycles for the first time. Under the optimal conditions, the benzothiophenes could be transformed into the beta-arylbenzothiophenes, and the benzofurans gave only alpha-aryl-substituted products.

Extending pi-conjugation system with benzene: An effective method to improve the properties of benzodithiophene-based polymer for highly efficient organic solar cells

To obtain a polymer based on benzodithiophene (BDT) owning both a largely extended pi-conjugation system and a low-lying highest occupied molecular orbital (HOMO), a polymer (PBDTBzT-DTffBT) containing benzothienyl-substituted BDT is designed and synthesized. Compared with the polymer (PBDTT-DTffBT) based on thienyl-substituted BDT, PBDTBzT-DTffBT exhibits better thermal stabilities, red-shifted absorption spectra, and stronger intermolecular interactions. The HOMO and lowest unoccupied molecular orbital (LUMO) in PBDTBzT-DTffBT are decreased by 0.11 and 0.13 eV, respectively, which should be attributed to the contribution of the electron-withdrawing group benzene. Polymer solar cells (PSCs) based on PBDTBzT-DTffBT and [6,6]-phenyl-C61-butyric acid methyl ester (PC61BM) exhibit a maximum power conversion efficiency (PCE) of 7.30% with a large open-circuit voltage of 0.90 V under AM 1.5G illumination (100 mW/cm2). The PCE is 36% higher than that of the PSCs derived from PBDTT-DTffBT. These findings provide a new approach to design high-performance conjugated polymers for efficient solution-processed PSCs.

Benzo[b]thiophenesulfonamide 1,1-dioxide derivatives (BTS) were described as candidate antineoplastic drugs. In the hope of finding new compounds with improved antitumour activity and reduced toxicity, we have designed and synthesized a small series of benzo[b]thiophene-6-carboxamide 1,1-dioxide derivatives (BTC) structurally related with the best reported BTS. Growth inhibition of HTB-54, CCRF-CEM and HeLa tumour cells lines at nanomolar concentrations was exhibited by some of the BTC. Hydrophobic substituents on the carboxamide group increased cytotoxicity but substitution by a hydroxy group diminished it, thus pointing to the electronic density on benzo[b]thiophene nucleus as a determinant factor. The process of cell death induced by BTC derivatives was further analyzed in CCRF-CEM cells, where these compounds induced apoptosis in a time and dose-dependent manner and cell cycle arrest at S phase. BTC derivatives also induced a significant increase in intracellular ROS levels in this cell line. Previous treatment of the cells with the antioxidant N-acetyl-cysteine abrogated the induction of apoptosis by BTC indicating that ROS generation is a previous event required to trigger the BTC induced apoptotic process.

4-Aryl Pyrrolidines as a Novel Class of Orally Efficacious Antimalarial Agents. Part 1: Evaluation of 4-Aryl- N-benzylpyrrolidine-3-carboxamides

Identification of novel chemotypes with antimalarial efficacy is imperative to combat the rise of Plasmodium species resistant to current antimalarial drugs. We have used a hybrid target-phenotype approach to identify and evaluate novel chemotypes for malaria. In our search for drug-like aspartic protease inhibitors in publicly available phenotypic antimalarial databases, we identified GNF-Pf-4691, a 4-aryl-N-benzylpyrrolidine-3-carboxamide, as having a structure reminiscent of known inhibitors of aspartic proteases. Extensive profiling of the two terminal aryl rings revealed a structure-activity relationship in which relatively few substituents are tolerated at the benzylic position, but the 3-aryl position tolerates a range of hydrophobic groups and some heterocycles. Out of this effort, we identified (+)-54b (CWHM-1008) as a lead compound. 54b has EC50 values of 46 and 21 nM against drug-sensitive Plasmodium falciparum 3D7 and drug-resistant Dd2 strains, respectively. Furthermore, 54b has a long half-life in mice (4.4 h) and is orally efficacious in a mouse model of malaria (qd; ED99 ? 30 mg/kg/day). Thus, the 4-aryl-N-benzylpyrrolidine-3-carboxamide chemotype is a promising novel chemotype for malaria drug discovery.
