Design and synthesis of Rho kinase inhibitors (II)

In a previous study, we identified several structurally unrelated scaffolds of the Rho kinase inhibitor using pharmacophore information obtained from the results of a high-throughput screening and structural information from a homology model of Rho kinase. 1H-Indazole is one of the candidate scaffolds on which a new series of potent Rho kinase inhibitors could be developed. In this study, the detailed structure-activity relationship of 1H-indazole analogues was studied. During this study, we found that the cell-free enzyme inhibitory potential of Rho kinase inhibitors having the 1H-indazole scaffold did not necessarily correlate with their inhibitory potential toward the chemotaxis of cultured cells. The choice of the linker substructure was shown to be an important factor for the 1H-indazole analogues to inhibit the chemotaxis of cells. Optimization of the 1H-indazole inhibitors with respect to the in vitro inhibition of monocyte chemotaxis induced by MCP-1 was carried out. The inhibitory potential was improved both in the cell-free enzyme assay and in the chemotaxis assay.

The crystal and molecular structures of two methoxybenzo[b]thiophenes have been determined by three-dimensional, single-crystal X-ray diffractometry. Both 3-(4?-hydroxy-3?,5?-dimethoxybenzoyl)-2-(4?- methoxyphenyl)-6-methoxybenzo[b]thiophene and 3-(2?,6?-dimethoxybenzoyl)-2-(4?-methoxyphenyl)-6- methoxybenzo[b]thiophene (hereafter referred to as I and II, respectively) crystallize in the monoclinic centrosymmetric space group P21/n (No. 14, C52h) with four formula units-per cell with a = 6.866(1), b = 28.638(2), c = 11.830(2) A, and beta = 105.52(1) and a = 9.328(1), b = 7.977(1), c = 29.650(4) A, and beta = 97.87(1), respectively. The phase problems were solved by direct methods and the respective final full-matrix least-squares refinements converged to R = 0.046 and 0.031. The structures differ in the positioning of the dimethoxy groups of the benzoyl ligands and the addition of a hydroxyl group in I. The molecules in the crystal lattice are held together by van der Waals forces plus the addition of hydrogen bonding in compound I. Selected bond distances and angles and torsion angles are tabularized.

Rational design of a visible-light photochromic diarylethene: a simple strategy by extending conjugation with electron donating groups

Photochromic diarylethenes have been widely used in many fields. However, their cyclization process must be induced by UV light. In this article, a simple strategy is developed by extending pi-conjugation with electron donating groups. The modified dirylethene derivative can photocyclolize under 405-nm light with a good photochromic efficiency. Meanwhile, its absorption and moderate fluorescence can be switched effectively in both directions by visible lights (405 and 520 nm, respectively) in different solutions and in living cells. We believe that this simple method will become a versatile strategy for developing various dirylethylenes with visible-light photochromism.

Oxidation mechanism of dibenzothiophene compounds: A computational study

The potential energy surfaces of the oxidation of three model heterocyclic organic sulfides dibenzothiophene (DBT), 4-methylbenzothiophene (MDBT) and 4,6-dimethyldibenzothiophene (2MDBT) were examined using HCO3H as oxidants adopting MP2, omegaB97X-D, M06-2X and B3LYP methods at 6-311+G (d,p) level. The stationary points on the potential energy surfaces for the first and second oxidation reaction were fully optimized and characterized. The natural orbital population analysis (NPA) was also performed to understand the charge distribution. The calculated results may contribute to understanding the reaction mechanism of the ODS of refractory DBT sulfides.

Herein we report on the synthesis and acid-responsive emission properties of donor-acceptor (D-A) molecules that contain a thienothiophene unit. 2-Arylthieno[3,2-b]thiophenes were conjugated with an N-methylbenzimidazole unit to form acid-responsive D-A-type fluorophores. The D-A-conjugated fluorophores showed intense intramolecular charge-transfer (ICT) emission in response to acid. The effect of the substitution on their photophysical properties as well as their solvent-dependence indicated non-twisting ICT emission in protonated D-A molecules. The quinoidal character of 2-arylthienothiophene as a donor part is discussed, as it is assumed that it contributes to suppression of the molecular twisting in the excited state, therefore decreasing the nonradiative rate constant, thereby resulting in the intense ICT emission. Acid-base-sensitive triple-color emission was also achieved by the introduction of a base-responsive phenol group in the donor part.

Catalytic Diastereo- and Enantioselective Fluoroamination of Alkenes

The stereoselective synthesis of syn-beta-fluoroaziridine building blocks via chiral aryl iodide-catalyzed fluorination of allylic amines is reported. The method employs HF-pyridine as a nucleophilic fluoride source together with mCPBA as a stoichiometric oxidant, and affords access to arylethylamine derivatives featuring fluorine-containing stereocenters in high diastereo- and enantioselectivity. Catalyst-controlled diastereoselectivity in the fluorination of chiral allylic amines enabled the preparation of highly enantioenriched 1,3-difluoro-2-amines bearing three contiguous stereocenters. The enantioselective catalytic method was applied successfully to other classes of multifunctional alkene substrates to afford anti-beta-fluoropyrrolidines, as well as a variety of 1,2-oxyfluorinated products.
